In humans over 15% of X‐linked genes have been shown to ‘escape’ from X‐chromosome inactivation (XCI): they continue to be expressed to some extent from the inactive X chromosome. Mono‐allelic expression is anticipated within a cell for genes subject to XCI, but random XCI usually results in expression of both alleles in a cell population. Using a study of allelic expression from cultured lymphoblasts and fibroblasts, many of which showed substantial skewing of XCI, we recently reported that the expression of (...) genes lies on a contiunuum between those that are subject to inactivation, and those that escape. We now review allelic expression studies from mouse, and discuss the variability in escape seen in both humans and mice in genic expression levels, between X chromosomes and between tissues. We also discuss current knowledge of the heterochromatic features, DNA elements and three‐dimensional topology of the inactive X that contribute to the balance of expression from the otherwise inactive X chromosome. Trust has been identified as an indicator within Social Quality theory. As an important component of social quality, trust has become increasingly important in modern society because literature suggests that trust in a number of democratic countries is declining. Modern technologies and specialties are often beyond the understanding of lay individuals and thus, the need for trusting relations between lay individuals and organizations/individuals has grown. The purpose of the study was to examine the extent to which Australians (dis)trust individuals and (...) organizations/institutions. A national postal survey was conducted with 1,044 respondents recruited using the electronic white pages directory. Findings from multivariate analyses suggest that income, age, sex, and health status are associated with trust in groups of individuals and trust in organizations/institutions. The findings highlight populations where trust needs to be (re)built. Future government policy and practice should utilize these findings as a means of facilitating social quality. (shrink)

In attempting to provide protection to individuals and communities, childhood immunization has benefits that far outweigh disease risks. However, some parents decide not to immunize their children with some or all vaccines for reasons including lack of trust in governments, health professionals, and vaccine manufacturers. This article employs a theoretical analysis of trust and distrust to explore how twenty-seven parents with a history of vaccine rejection in two Australian cities view the expert systems central to vaccination policy and practice. Our (...) data show how perceptions of the profit motive generate distrust in the expert systems pertaining to vaccination. Our participants perceived that pharmaceutical companies had a pernicious influence over the systems driving vaccination: research, health professionals, and government. Accordingly, they saw vaccine recommendations in conflict with the interests of their child and “the system” underscored by malign intent, even if individual representatives of this system were not equally tainted. This perspective was common to parents who declined all vaccines and those who accepted some. We regard the differences between these parents—and indeed the differences between vaccine decliners and those whose Western medical epistemology informs reflexive trust—as arising from the internalization of countering views, which facilitates nuance. Using telephone interviews among a random sample of 537 Dutch children aged 7–12 years old, we investigated (a) the prevalence of fear reactions to television news among younger and older children and among boys and girls, (b) what types of news items children in different age and gender groups refer to as frightening, and (c) whether children's fear reactions to regular adult television news differed from their fear reactions to a special children's news program. Overall, 48.2 % of the children (...) who reported watching the adult or children's news programs, reported fear reactions to adult news, while 32.6 % reported fear of children's news content. For both types of news programs, younger children and girls more often reported fear than older children and boys did. The most prevalent fear-inducing news content categories were (a) interpersonal violence, (b) fires, accidents, and disasters, and (c) visual depictions of the consequences of violence. (shrink)

-/- It is commonly believed that the etiology of autism is at least partly explained through genetics. Given the complexity of autism and the variability of the autistic phenotype, genetic research and counseling in this field are also complex and associated with specific ethical questions. Although the ethics of autism genetics, especially with regard to reproductive choices, has been widely discussed on the public fora, an in depth philosophical or bioethical reflection on all aspects of the theme seems to be (...) missing. With this literature review we wanted to map the basic questions and answers that exist in the bioethical literature on autism genetics, research, counseling and reproduction, and provide suggestions as to how the discussion can proceed. We found 19 papers that fitted the description of “bioethics literature focusing on autism genetics,” and analyzed their content to distill arguments and themes. We concluded that because of the complexity of autism, and the uncertainty with regard to its status, more ethical reflection is needed before definite conclusions and recommendations can be drawn. Moreover, there is a dearth of bioethical empirical studies querying the opinions of all parties, including people with autism themselves. Such empirical bioethical studies should be urgently done before bioethical conclusions regarding the aims and desirability of research into autism genes can be done. Also, fundamental philosophical reflection on concepts of disease should accompany research into the etiology of autism. © 2016 Wiley Periodicals, Inc. (shrink)

The paper examines the problem of textual/cultural dynamics linked to the issue of semiotic literariness, to be further investigated by the authors in later papers on literary semiotics. This scientific project aims to get closer to reaching an adequate disciplinary identification for semiotics of literature and a relatively precise definition of the status of this field in relation to semiotics of culture. The first step for the project is to reveal the interrelationhip between text and culture using the notion of (...) dynamics that can be reconstructed from a historical perspective through some essential components of Formalist and Structuralist theory (Tynyanov’s ‘function’, Jakobson’s ‘dominant’) and also works by Lotman (the ‘text–culture’ relationship) and Bakhtin (‘dialogue’). The notions of inclusiveness/integration, distancing and hierarchization, leading to transformation, are interpreted in some detail in the context of these theories. On these grounds, three basic categories of the analysability of textual/cultural dynamics are set up with the indication of further aspects of the dynamic function: (1) mediation; (2) transposition; (3) temporality–spatiality. The suggested classification and the implied conceptual segmentation are expected to contribute to a synthesis between “Structuralist” and Peircean theoretical and methodological orientations in semiotic literary studies. This also reveals the need for a coexistence of approaches (a) moving from particular cultural fields (literary culture tradition) towards general semiotics of culture, and (b) returning from universal transfield concepts to literary culture, including the historical traditions both in art (object-level) and its scientific interpretation (meta-level). Prefrontal cortex is dysregulated in women with restricting anorexia nervosa. It is not known whether appetitive non-conscious stimuli bias cognitive responses in those with RAN. Thirteen women with RAN and 20 healthy controls completed a dorsolateral PFC working memory task and an anterior cingulate cortex conflict task, while masked subliminal food, aversive and neutral images were presented. During the DLPFC task, accuracy was higher in the RAN compared to the HC group, but superior performance was compromised when subliminal food stimuli (...) were presented: errors positively correlated with self-reported trait anxiety in the RAN group. These effects were not observed in the ACC task. Appetitive activation is intact and anxiogenic in women with RAN, and non-consciously interacts with working memory processes associated with the DLPFC. This interaction mechanism may underlie cognitive inhibition of appetitive processes that are anxiety inducing, in people with AN. (shrink)

This study was used to evaluate whether the proximity of dogs to their human companions during sleep is associated with common problematic behaviors in canines, such as destroying objects, vocalizing excessively, urinating/defecating in inappropriate places (all when dogs are alone at home), and aggressive threats (growling or barking) and acts (biting) toward people. Over 60,000 dog keepers answered an online questionnaire that addressed where their dogs slept at night and the frequency with which they exhibited such behaviors. Except urinating/defecating in (...) inappropriate places and biting people, other problematic behaviors were less frequent in dogs who slept inside the house. We conclude that dogs sleeping indoors (that is, closer to their keepers) less frequently exhibit aggressive threats and problematic behaviors that are commonly associated with separation anxiety. The Dutch policy objective of a fully sustainable livestock sector without mutilations by 2023 is not compatible with the routine practice of tail docking to minimize the risk of tail biting. To examine farmer attitudes towards docking, a telephone survey was conducted among 487 conventional and 33 organic Dutch pig farmers. “Biting” (of tails, ears, or limbs) was identified by the farmers as a main welfare problem in pig farming. About half of the farmers reported to have no tail biting (...) problems in their own herd. When farmers did report problems, they most often reported figures between 1 and 5 % of the animals. High incidences of tail biting were anticipated when trying to keep undocked pigs. Enrichment materials used in the conventional sector included mainly chains (52–63 % of the farms) and hanging rubber or plastic balls (22–30 %). Straw, sawdust, or wood shavings was hardly provided in conventional pig farming (2–3 %), in contrast to organic farming (88–100 % of farms). Conventional pig farmers feel a curly tail is not very important for sustainable pig farming. They consider enrichment to be less effective and tail docking to be less stressful for them and their piglets than their organic colleagues do. Pig farmers identified climate as a main risk factor for tail biting as opposed to enrichment. The objective of reducing routine tail docking requires solutions for dealing with tail biting problems at the farm level. In this process, transfer of scientific knowledge about enrichment materials and other measures to prevent and cure tail biting is critical, as is a change in farmer attitudes and awareness of the moral issues involved. Eugene Hargrove’s impact on the field of environmental philosophy is hard to adequately convey. His service as an activist, educator, and the founder of the journal Environmental Ethics created the context in which the field thrived. His legacy also provides a strong foundation for environmental ethics and activism of the future. Today, environmental philosophy is expanding to include human-dominated environments, once thought to fall outside of the purview of the discipline. Work in these “blind spots” is imperative if we wish (...) to improve sustainability and stem the ecological bleeding that marks the Anthropocene. Hargrove’s theoretical work on “weak anthropocentric intrinsic value” opens a space for environmental ethics to better understand human-dominated environments. If Paul B. Thompson is right that the way we farm should be of utmost concern for environmentalists, then Hargrove’s insights could help protect large swaths of land beyond parkland borders. (shrink)

What happens when the expression of parental values in child rearing runs contrary to the claim of children to be loved and respected by parents? This chapter asks whether parents who hold, and seek to express, attitudes and beliefs that are contemptuous of sexual minorities are competent parents. We argue that homophobic views held by parents can pose a serious threat to the well-being of children and that adults who harbour such views fall below the threshold of competency requisite to (...) acquiring and maintaining the moral right to parent. We explore this issue by considering whether conservative religious fundamentalists who hold and express strongly homophobic beliefs should be considered competent parents. Some religious fundamentalists seem unwilling to love and respect children who are or might be gay. The systematic denial of love and respect to gay youth by homophobic parents causes a great deal of harm to children and arguably constitutes a form of abuse. The basic structure of the argument we set out is simple: (a) children have a right to affective caring; (b) competent prospective parents must be able to reliably provide affective caring to children; but (c) strongly homophobic parents cannot reliably provide affective to caring to children who are gay; so (d) strongly homophobic adults are not competent parents. -/- . Food and farming are research topics of marked complexity. His expansive research portfolio carves out a unique space in the discipline, as it acts as a bridge between philosophical subfields and beyond, engaging deeply with a surprising range of scientific disciplines. The development of narrative skill has been investigated extensively in a wide range of languages, cross-linguistically and in multilingual settings (Berman & Slobin, 1994b; Hickmann, 2004; Severing & Verhoeven 2001, Strömqvist & Verhoeven, 2004). The present study investigates the development of reference realisation in narrative among Indigenous children in a remote urban township in Central Australia. The children, aged between 5 and 14 years, are speakers of a contact language, Wumpurrarni English. Language development is rarely investigated among speakers of minority (...) languages, whose language development is often appraised in the majority language, with little attention to language performance in the speaker’s home variety. The present study addresses this gap through a fine-grained qualitative analysis of the development of reference in narrative, drawing on a complex stimulus and a model of discourse strategy. The results show a) a developmental trajectory similar to that found in other languages, with children aged eight and under producing simpler and less globally organised narratives than older speaker groups, and b) vulnerability to the changing demands of the stimulus among these younger speakers. In addition, a subset of narrations were produced in ‘school variety’, a style more like Standard Australian English, and the results for this set showed that the narrative content and global organisation of the productions by ten- and twelve–year-olds was more similar to the productions of younger children, than like-aged speakers narrating in their home variety. Analysis of speaker responses to two factors of complexity, the stimulus and code choice, illuminated mechanisms for discourse production and development, and suggest that constructing discourse requires co-ordination of an underlying schema and on-line construction of a particular story through the deployment of linguistic devices in a particular narrative context, and that these two skills are tightly interdependent, and indeed co-constructing. This book review evaluates Kenneth B. Kidd's Theory for Beginners: Children's Literature as Critical Thought, focusing specifically on Kidd's exploration of the symbiotic relationship between children's literature, philosophy, and critical theory.

Pericytes, like vascular smooth muscle cells, are perivascular cells closely associated with blood vessels throughout the body. Pericytes are necessary for vascular development and homeostasis, with particularly critical roles in the brain, where they are involved in regulating cerebral blood flow and establishing the blood-brain barrier. A role for pericytes during neurovascular disease pathogenesis is less clear—while some studies associate decreased pericyte coverage with select neurovascular diseases, others suggest increased pericyte infiltration in response to hypoxia or traumatic brain injury. Here, (...) we used an endothelial loss-of-function Recombination signal binding protein for immunoglobulin kappa J region (Rbpj)/Notch mediated mouse model of brain arteriovenous malformation (AVM) to investigate effects on pericytes during neurovascular disease pathogenesis. We tested the hypothesis that pericyte expansion, via morphological changes, and Platelet-derived growth factor B/Platelet-derived growth factor receptor β (Pdgf-B/Pdgfrβ)-dependent endothelial cell-pericyte communication are affected, during the pathogenesis of Rbpj mediated brain AVM in mice. Our data show that pericyte coverage of vascular endothelium expanded pathologically, to maintain coverage of vascular abnormalities in brain and retina, following endothelial deletion of Rbpj. In Rbpj-mutant brain, pericyte expansion was likely attributed to cytoplasmic process extension and not to increased pericyte proliferation. Despite expanding overall area of vessel coverage, pericytes from Rbpj-mutant brains showed decreased expression of Pdgfrβ, Neural (N)-cadherin, and cluster of differentiation (CD)146, as compared to controls, which likely affected Pdgf-B/Pdgfrβ-dependent communication and appositional associations between endothelial cells and pericytes in Rbpj-mutant brain microvessels. By contrast, and perhaps by compensatory mechanism, endothelial cells showed increased expression of N-cadherin. Our data identify cellular and molecular effects on brain pericytes, following endothelial deletion of Rbpj, and suggest pericytes as potential therapeutic targets for Rbpj/Notch related brain AVM. As improvements in neuroscience have enabled a better understanding of disorders of consciousness as well as methods to treat them, a hurdle that has become all too prevalent is the denial of coverage for treatment and rehabilitation services. In 2011, a settlement emerged from a Vermont District Court case, Jimmo v. Sebelius, which was brought to stop the use of an “improvement standard” that required tangible progress over an identifiable period of time for Medicare coverage of services. While the use (...) of this standard can have deleterious effects on those with many chronic conditions, it is especially burdensome for those in the minimally conscious state, where improvements are unpredictable and often not manifested through repeatable overt behaviors. Though the focus of this paper is on the challenges of brain injury and the minimally conscious state, which an estimated 100,000 to 200,000 individuals suffer from in the United States, the post-Jimmo arguments presented can and should have a broad impact as envisioned by the plaintiffs who brought the case on behalf of multiple advocacy groups representing patients with a range of chronic care conditions. Whole–exome sequencing (WES) has increased our ability to analyze large parts of the human genome, bringing with it a plethora of ethical, legal, and social implications. A topic dominating discussion of WES is identification of “secondary findings” (SFs), defined as the identification of risk in an asymptomatic individual unrelated to the indication for the test. SFs can have considerable psychosocial impact on patients and families, and patients with an SF may have concerns regarding genomic privacy and genetic discrimination. The Genetic (...) Information Nondiscrimination Act of 2008 (GINA) currently excludes protections for members of the military. This may cause concern in military members and families regarding genetic discrimination when considering genetic testing. In this report, we discuss a case involving a patient and family in which a secondary finding was discovered by WES. The family members have careers in the U.S. military, and a risk–predisposing condition could negatively affect employment. While beneficial medical management changes were made, the information placed exceptional stress on the family, who were forced to navigate career–sensitive “extra–medical” issues, to consider the impacts of uncovering risk–predisposition, and to manage the privacy of their genetic information. We highlight how information obtained from WES may collide with these issues and emphasize the importance of genetic counseling for anyone undergoing WES. (shrink)

The aim of this chapter is to elucidate Kant’s account of the a priori exercise of imagination in the B-edition of the Transcendental Deduction. It focuses, in particular, on issues surrounding the relationship that the a priori exercise of imagination has to space and time in light of Kant’s distinction between ‘formal intuitions’ and ‘forms of intuition’. The chapter argues against a radical reading of Kant, according to which the a priori exercise of imagination is the source of our most (...) fundamental pure intuitions of space and time that condition empirical intuitions. It, instead, defends a mediating model that is in keeping with Kant’s theory of imagination more generally. According to this mediating model, the a priori exercise of imagination mediates between pure intuitions of space and time and the categories, and it makes possible the way we take up empirical intuitions given through sense in perception and experience. Samantha Vice’s proposal on how to live in ‘this strange place’ of contemporary South Africa, includes an appeal to the concepts of shame and silence. In this paper, I use Emmanuel Levinas and Giorgio Agamben to move the discussion of shame from a moral to an existential question. The issue is not about how one should feel, but about the kind of self that whiteness in South Africa makes possible today. Shame desubjectifies. Vice’s recommendation of silence is then taken (...) as witnessing/listening, which I argue grounds the possibility of a recovery of the self. (shrink)

In the Critique of the Power of Judgment, Kant offers a theory of artistic expression in which he claims that a work of art is a medium through which an artist expresses an ‘aesthetic idea’. While Kant’s theory of aesthetic ideas often receives rather restrictive interpretations, according to which aesthetic ideas can either present only moral concepts, or only moral concepts and purely rational concepts, in this article I offer an ‘inclusive interpretation’ of aesthetic ideas, according to which they can (...) present not only moral and purely rational concepts but also empirical concepts and emotions related to our ordinary experience. Although this latter class of experience-oriented aesthetic ideas has been neglected, I argue that recognizing the role it plays in Kant’s account is crucial for understanding his views not only of artistic production and our experience of art but also of the value he takes art to have for our ordinary experience of the world, others, and our own selves. What is more, insofar as the inclusive interpretation brings to light Kant’s acknowledgement of the close connection between experience and art, it reveals that his overall view of art is more plausible than is often thought, and recommends it as worthy of further consideration. It has become typical to read Kant and Merleau-Ponty as offering competing approaches to perceptual experience. Kant is interpreted as an ‘intellectualist’ who regards perception as conceptual ‘all the way out’, while Merleau-Ponty is seen as Kant’s challenger, who argues that perception involves non-conceptual, embodied ‘coping’. In this paper, however, I argue that a closer examination of their views of perception, especially with respect to the notion of ‘schematism’, reveals a great deal of historical and philosophical continuity between them. By (...) analyzing Kant’s theory of schematism, the interpretation of it by the Neo-Kantian Pierre Lachièze-Rey, and Merleau-Ponty’s theory of the body schema, we find that aspects of Merleau-Ponty’s theory of perception are better understood as a development of Kant’s theory of perception. In the Critique of Pure Reason, Kant describes schematism as a (A141/B180–1). While most commentators treat this as Kant's metaphorical way of saying schematism is something too obscure to explain, I argue that we should follow up Kant's clue and treat schematism literally as Kunst. By letting our interpretation of schematism be guided by Kant's theoretically exact ways of using the term Kunst in the Critique of Judgment we gain valuable insight into the nature of schematism, as well as its (...) connection to Kant's concerns in the third Critique. Thomas Hobbes is an iconic figure who serves as an easy reference for pundits commenting on the brutality of war as well as for critics of a distinctly modern individualism in which calculating and rapacious self-interest is the cause of the violence, destruction, and exploitation endemic to the contemporary world. Frost's reading of Hobbes's philosophy shows us that underlying such visions of self and politics is another iconic figure: that of the Cartesian subject. What gives the iconic Hobbes his hardcore (...) individualism and its corollary accounts of instrumentalism, conflict, and absolutism is a Cartesian rendering of the self as split into mind and body. Carefully elaborating Hobbes's materialist ontology, _Lessons from a Materialist Thinker_ challenges both our implicit Cartesian assumptions about the self and the commonplace Hobbes that so readily figures violence in our political imagination. Through his materialism, Hobbes presents an alternative modern account of self-consciousness, reason, agency, power, freedom, and responsibility. In doing so, he shows that our fundamental intersubjectivity and interdependence require that we pursue peace above all else. (shrink)
